Riding to Rochester, NY this summer

My 25 year old daughter and I are planning a trip to Rochester, NY. I'm thinking of the best routes that avoid many tough climbs, haha, starting point is from Howard county.
One way is to ride to Cumberland and take the GAP to Pittsburgh then roads to to Erie, PA.,then hug Lake Erie to Buffalo then the Erie Canal Towpath to Rochester. About 500 miles to my rough estimation.
or:
Bike to near Cumberland to Ellerslie, Md. at Rt.35 turns into Rt. 96 in PA. and pick up what PA. calls their Route G route "surprisingly flat"

on roads until you get to the Pine Creek Trail (enter at rt. 414 at Hilborn and exit at SR 362 Darling Run, looks to be about 40 or so miles on the crushed gravel trail. Then back to roads pass by Mansfield until the NY border.
Now from here I have no idea which roads to take to Rochester that would include the fewest mountains, climbs etc.
